Modeling- and Simulation-Driven Methodology for the Deployment of an Inland Water Monitoring System

In response to the challenges introduced by global warming and increased eutrophication, this paper presents an innovative modeling and simulation (M&S)-driven model for developing an automated inland water monitoring system. This system is grounded in a layered Internet of Things (IoT) architecture and seamlessly integrates cloud, fog, and edge computing to enable sophisticated, real-time environmental surveillance and prediction of harmful algal and cyanobacterial blooms (HACBs). Utilizing autonomous boats as mobile data collection units within the edge layer, the system efficiently tracks algae and cyanobacteria proliferation and relays critical data upward through the architecture. These data feed into advanced inference models within the cloud layer, which inform predictive algorithms in the fog layer, orchestrating subsequent data-gathering missions. This paper also details a complete development environment that facilitates the system lifecycle from concept to deployment. The modular design is powered by Discrete Event System Specification (DEVS) and offers unparalleled adaptability, allowing developers to simulate, validate, and deploy modules incrementally and cutting across traditional developmental phases.

Microcystin-LR Induces Estrogenic Effects at Environmentally Relevant Concentration in Black-Spotted Pond Frogs (Pelophylax nigromaculatus): In Situ, In Vivo, In Vitro, and In Silico Investigations.

Harmful cyanobacterial blooms are frequent and intense worldwide, creating hazards for aquatic biodiversity. The potential estrogen-like effect of Microcystin-LR (MC-LR) is a growing concern. In this study, we assessed the estrogenic potency of MC-LR in black-spotted frogs through combined field and laboratory approaches. In 13 bloom areas of Zhejiang province, China, the MC-LR concentrations in water ranged from 0.87 to 8.77 μg/L and were correlated with sex hormone profiles in frogs, suggesting possible estrogenic activity of MC-LR. Tadpoles exposed to 1 μg/L, an environmentally relevant concentration, displayed a female-biased sex ratio relative to controls. Transcriptomic results revealed that MC-LR induces numerous and complex effects on gene expression across multiple endocrine axes. In addition, exposure of male adults significantly increased the estradiol (E2)/testosterone (T) ratio by 3.5-fold relative to controls. Downregulation of genes related to male reproductive endocrine function was also identified. We also showed how MC-LR enhances the expression of specific estrogen receptor (ER) proteins, which induce estrogenic effects by activating the ER pathway and hypothalamic-pituitary-gonadal (HPG) axis. In aggregate, our results reveal multiple lines of evidence demonstrating that, for amphibians, MC-LR is an estrogenic endocrine disruptor at environmentally relevant concentrations. The data presented here support the need for a shift in the MC-LR risk assessment. While hepatoxicity has historically been the focus of MC-LR risk assessments, our data clearly demonstrate that estrogenicity is a major mode of toxicity at environmental levels and that estrogenic effects should be considered for risk assessments on MC-LR going forward.

Profiling the interplay and coevolution of Microcystis aeruginosa and cyanosiphophage Mic1.

The highly polymorphic Microcystis aeruginosa is one of the predominant bloom-forming cyanobacteria in eutrophic freshwater bodies and is infected by diverse and abundant cyanophages. The presence of a large number of defense systems in M. aeruginosa genome suggests a dynamic interplay and coevolution with the cyanophages. In this study, we investigated the temporal gene expression pattern of Mic1 after infection and the corresponding transcriptional responses of its host. Moreover, the identification of a less-infective Mic1 and a Mic1-resistant host strain provided the evolved genes in the phage-host coevolution during the multiple-generation cultivation in the laboratory. Our findings enrich the knowledge on the interplay and coevolution of M. aeruginosa and its cyanophages and lay the foundation for the future application of cyanophage as a potential eco-friendly and bio-safe agent in controlling the succession of harmful cyanobacterial blooms.

Cyanotoxin cylindrospermopsin disrupts lipid homeostasis and metabolism in a 3D in vitro model of the human liver

Cylindrospermopsin, a potent hepatotoxin produced by harmful cyanobacterial blooms, poses environmental and human health concerns. We used a 3D human liver in vitro model based on spheroids of HepG2 cells in combination with molecular and biochemical assays, automated imaging, targeted LC-MS-based proteomics, and lipidomics to explore cylindrospermopsin effects on lipid metabolism and the processes implicated in hepatic steatosis. Cylindrospermopsin (1 μM, 48 h) did not significantly affect cell viability but partially reduced albumin secretion. However, it increased neutral lipid accumulation in HepG2 spheroids while decreasing phospholipid levels. Simultaneously, cylindrospermopsin upregulated genes for lipogenesis regulation (SREBP1) and triacylglycerol synthesis (DGAT1/2) and downregulated genes for fatty acid synthesis (ACLY, ACCA, FASN, SCD1). Fatty acid uptake, oxidation, and lipid efflux genes were not significantly affected. Targeted proteomics revealed increased levels of perilipin 2 (adipophilin), a major hepatocyte lipid droplet-associated protein. Lipid profiling quantified 246 lipid species in the spheroids, with 28 significantly enriched and 15 downregulated by cylindrospermopsin. Upregulated species included neutral lipids, sphingolipids (e.g., ceramides and dihexosylceramides), and some glycerophospholipids (phosphatidylethanolamines, phosphatidylserines), while phosphatidylcholines and phosphatidylinositols were mostly reduced. It suggests that cylindrospermopsin exposures might contribute to developing and progressing towards hepatic steatosis or metabolic dysfunction-associated fatty liver disease (MAFLD).

Analytical Methods for Anatoxin-a Determination: A Review.

Anatoxin-a (ATX-a) is a potent neurotoxin produced by several species of cyanobacteria whose exposure can have direct consequences, including neurological disorders and death. The increasing prevalence of harmful cyanobacterial blooms makes the detection and reliable assessment of ATX-a levels essential to prevent the risk associated with public health. Therefore, the aim of this review is to compile the analytical methods developed to date for the detection and quantification of ATX-a levels alone and in mixtures with other cyanotoxins and their suitability. A classification of the analytical methods available is fundamental to make an appropriate choice according to the type of sample, the equipment available, and the required sensitivity and specificity for each specific purpose. The most widely used detection technique for the quantification of this toxin is liquid chromatography-tandem mass spectrometry (LC-MS/MS). The analytical methods reviewed herein focus mainly on water and cyanobacterial samples, so the need for validated analytical methods in more complex matrices (vegetables and fish) for the determination of ATX-a to assess dietary exposure to this toxin is evidenced. There is currently a trend towards the validation of multitoxin methods as opposed to single-ATX-a determination methods, which corresponds to the real situation of cyanotoxins' confluence in nature.

Fungal removal of cyanotoxins in constructed wetlands: The forgotten degraders

Harmful cyanobacterial blooms have increased globally, releasing hazardous cyanotoxins that threaten the safety of water resources. Constructed wetlands (CWs) are a nature-based and low-cost solution to purify and remove cyanotoxins from water. However, bio-mechanistic understanding of the biotransformation processes expected to drive cyanotoxin removal in such systems is poor, and primarily focused on bacteria. Thus, the present study aimed at exploring the fungal contribution to microcystin-LR and cylindrospermopsin biodegradation in CWs. Based on CW mesocosms, two experimental approaches were taken: a) amplicon sequencing studies were conducted to investigate the involvement of the fungal community; and b) CW fungal isolates were tested for their microcystin-LR and cylindrospermopsin degradation capabilities. The data uncovered effects of seasonality (spring or summer), cyanotoxin exposure, vegetation (unplanted, Juncus effusus or Phragmites australis) and substratum (sand or gravel) on the fungal community structure. Additionally, the arbuscular mycorrhizal fungus Rhizophagus and the endophyte Myrmecridium showed positive correlations with cyanotoxin removal. Fungal isolates revealed microcystin-LR-removal potentials of approximately 25 % in in vitro biodegradation experiments, while the extracellular chemical fingerprint of the cultures suggested a potential intracellular metabolization. The results from this study may help us understand the fungal contribution to cyanotoxin removal, as well as their ecology in CWs.

Inhibitory effect of malonic acid on Microcystis aeruginosa: Cell morphology, oxidative stress and gene expression

The utilization of allelochemicals, which are regarded as eco-friendly algaecides for mitigating harmful cyanobacterial blooms (HCB), has attracted increasing attention, owing to their negligible toxicity, high selectivity and excellent degradability. In the present study, the inhibitory effects of malonic acid on Microcystis aeruginosa were investigated in terms of growth indicators, cell morphology, antioxidant enzymes activities and gene expression. The results indicated that the exposure to 0.5 mmol L−1 malonic acid for 7 d caused a high microalgae inhibition rate of 86.88 %. Furthermore, the analysis of scanning electron microscopy (SEM) and transmission electron microscope (TEM) showed that the presence of malonic acid resulted in the disruption of cell integrity and the changes in cell morphology. Specifically, malonic acid was observed to cleave microalgae cells, while concurrently oxidizing the microalgae organic matter. In addition, under the exposure of 0.5 mmol L−1 malonic acid, the levels of malondialdehyde (MDA) were 3.28 times as the control group on d-5, while the activities of superoxide dismutase (SOD) and catalase (CAT) were first increased by 5.96 and 11.73 fold compare to the control group on d-5, and then decreased, due to oxidative stress induced by malonic acid. Transcriptome-based gene expression analysis revealed that the presence of malonic acid significantly inhibited the transcription of genes (rbcL, psbA1) associated with photosynthesis, and the expression of DNA repair gene (recA), stress response gene (ftsH) and microcystin synthesis genes (mcyD, mcyH) were up-regulated. These findings provide novel insights into the inhibitory effect of malonic acid on M. aeruginosa, suggesting that malonic acid has the potential to control M. aeruginosa blooms.

Capture and flocculation of toxic cyanobacteria by amphiphilic peptide dendrimers for mitigating harmful blooms

Harmful and toxic cyanobacterial blooms in freshwater constitute critical environmental problems and are becoming more frequent as a consequence of global climate change. One mitigation strategy is flocculation, which could be achieved with treatment by highly efficient, non-toxic, and biodegradable agents. Here, we report two amphiphilic peptide dendrimers (AmPDs), KK2 and KK2K4, for the removal of the toxic and bloom-forming cyanobacterium Microcystis aeruginosa from freshwater. KK2 and KK2K4 are composed of a hydrophobic alkyl chain and a positively charged polylysine dendron of the first and second generation, respectively. Owing to electrostatic interactions with the negatively charged cell surface of M. aeruginosa, KK2 and KK2K4 could quickly capture the cyanobacterial cell population via flocculation at very low concentrations. When combined with the natural clay sepiolite, the two AmPDs were even more efficient in promoting cell flocculation. The most performant system, with 3.0 mg/L KK2K4 and sepiolite clay, could remove 97.1 % of M. aeruginosa within 15 min by forming stable flocs. Remarkably, such a system was able to trap toxin molecules released by cyanobacterial cells, thus limiting its destructive impact on ecosystems. This study demonstrates how self-assembling dendrimer materials can resolve cyanobacterial blooms, providing an innovative solution to this environmental challenge.

Eco-friendly management of harmful cyanobacterial blooms in eutrophic lakes through vertical flow multi-soil-layering technology

Eutrophication has led to the widespread occurrence of cyanobacterial blooms. Toxic cyanobacterial blooms with high concentrations of microcystins (MCs) have been identified in the Lalla Takerkoust reservoir in Morocco. The objective of this study was to evaluate the efficiency of the Multi-Soil-Layering (MSL) ecotechnology in removing natural cyanobacterial blooms from the lake. Two MSL pilots were used in rectangular glass tanks (60 × 10 × 70 cm). They consisted of permeable layers (PLs) made of pozzolan and a soil mixture layer (SML) containing local soil, ferrous metal, charcoal and sawdust. The main difference between the two systems was the type of local soil used: sandy soil for MSL1 and clayey soil for MSL2. Both MSL pilots effectively reduced cyanobacterial cell concentrations in the treated water to very low levels (0.09 and 0.001 cells/mL). MSL1 showed a gradual improvement in MC removal from 52 % to 99 %, while MSL2 started higher at 90 % but dropped to 54% before reaching 86%. Both MSL systems significantly reduced organic matter levels (97.2 % for MSL1 and 95.8 % for MSL2). Both MSLs were shown to be effective in removing cyanobacteria, MCs, and organic matter with comparable performance.

Biological and Chemical Approaches for Controlling Harmful Microcystis Blooms.

The proliferation of harmful cyanobacterial blooms dominated by Microcystis aeruginosa has become an increasingly serious problem in freshwater ecosystems due to climate change and eutrophication. Microcystis-blooms in freshwater generate compounds with unpleasant odors, reduce the levels of dissolved O2, and excrete microcystins into aquatic ecosystems, potentially harming various organisms, including humans. Various chemical and biological approaches have thus been developed to mitigate the impact of the blooms, though issues such as secondary pollution and high economic costs have not been adequately addressed. Red clays and H2O2 are conventional treatment methods that have been employed worldwide for the mitigation of the blooms, while novel approaches, such as the use of plant or microbial metabolites and antagonistic bacteria, have also recently been proposed. Many of these methods rely on the generation of reactive oxygen species, the inhibition of photosynthesis, and/or the disruption of cellular membranes as their mechanisms of action, which may also negatively impact other freshwater microbiota. Nevertheless, the underlying molecular mechanisms of anticyanobacterial chemicals and antagonistic bacteria remain unclear. This review thus discusses both conventional and innovative approaches for the management of M. aeruginosa in freshwater bodies.

Design, Synthesis, and Mode of Action of Thioacetamide Derivatives as the Algicide Candidate Based on Active Substructure Splicing Strategy.

Lakes and reservoirs worldwide are experiencing a growing problem with harmful cyanobacterial blooms (HCBs), which have significant implications for ecosystem health and water quality. Algaecide is an effective way to control HCBs effectively. In this study, we applied an active substructure splicing strategy for rapid discovery of algicides. Through this strategy, we first optimized the structure of the lead compound S5, designed and synthesized three series of thioacetamide derivatives (series A, B, C), and then evaluated their algicidal activities. Finally, compound A3 with excellent performance was found, which accelerated the process of discovering and developing new algicides. The biological activity assay data showed that A3 had a significant inhibitory effect on M. aeruginosa. FACHB905 (EC50 = 0.46 μM) and Synechocystis sp. PCC6803 (EC50 = 0.95 μM), which was better than the commercial algicide prometryn (M. aeruginosa. FACHB905, EC50 = 6.52 μM; Synechocystis sp. PCC6803, EC50 = 4.64 μM) as well as better than lead compound S5 (M. aeruginosa. FACHB905, EC50 = 8.80 μM; Synechocystis sp. PCC6803, EC50 = 7.70 μM). The relationship between the surface electrostatic potential, chemical reactivity, and global electrophilicity of the compounds and their activities was discussed by density functional theory (DFT). Physiological and biochemical studies have shown that A3 might affect the photosynthesis pathway and antioxidant system in cyanobacteria, resulting in the morphological changes of cyanobacterial cells. Our work demonstrated that A3 might be a promising candidate for the development of novel algicides and provided a new active skeleton for the development of subsequent chemical algicides.

Draft genome of novel cyanobacteria Sphaerothrix Gracilis isolated from coastal microplastics reveal insights to chemical ecology, bloom and plastic-utilization potential.

Cyanobacteria are oxygenic photosynthetic organisms which are found across many ecosystems, including freshwater and marine habitats. They are also found on natural and artificial surfaces. In this study, we cultured and characterise a novel cyanobacterium from the surfaces of foam microplastics of tropical coastal waters. We study the chemical ecology of this cyanobacterium, Sphaerothrix gracilis gen. et sp. nov., together with its potential to form harmful cyanobacterial blooms and bioremediation applications to combat plastic pollution. The genome of S. gracilis spanned 6.7 Mbp, with identification of antibiotic resistance, nitrogen-fixation, plastic-degrading and genes involved in harmful metabolite production. The transport of potentially harmful S. gracilis in coastal environments could have severe implications on human health and food security, especially in times of a cyanobacterial bloom.

Detection of Anatoxins in Human Urine by Liquid Chromatography Triple Quadrupole Mass Spectrometry and ELISA.

Harmful cyanobacterial blooms are becoming more common and persistent around the world. When in bloom, various cyanobacterial strains can produce anatoxins in high concentrations, which, unlike other cyanobacterial toxins, may be present in clear water. Potential human and animal exposures to anatoxins occur mainly through unintentional ingestion of contaminated algal mats and water. To address this public health threat, we developed and validated an LC-MS/MS method to detect anatoxins in human urine to confirm exposures. Pooled urine was fortified with anatoxin-a and dihydroanatoxin at concentrations from 10.0 to 500 ng/mL to create calibrators and quality control samples. Samples were diluted with isotopically labeled anatoxin and solvent prior to LC-MS/MS analysis. This method can accurately quantitate anatoxin-a with inter- and intraday accuracies ranging from 98.5 to 103% and relative standard deviations < 15%, which is within analytical guidelines for mass spectrometry methods. Additionally, this method qualitatively detects a common degradation product of anatoxin, dihydroanatoxin, above 10 ng/mL. We also evaluated a commercial anatoxin-a ELISA kit for potential diagnostic use; however, numerous false positives were detected from unexposed individual human urine samples. In conclusion, we have developed a method to detect anatoxins precisely and accurately in urine samples, addressing a public health area of concern, which can be applied to future exposure events.

A Bleeding Edge Web Application for Early Detection of Cyanobacterial Blooms

Harmful Algal and Cyanobacterial Bloom (HACB) threaten aquatic ecosystems, human health, and the economy. Many factors influence these dynamic events, which are often difficult to detect until the late stages of growth. The inclusion of an Early Warning System (EWS) can be instrumental in identifying hazards and preventing or at least minimizing their impact. Traditional monitoring approaches often fail to provide the real-time, high-resolution data needed for effective early warnings. The integration of Internet of Things (IoT) technologies offers a promising avenue to address these limitations by creating a network of interconnected sensors capable of continuously collecting and transmitting data from various aquatic environments. In this paper, we present DEVS-BLOOM-WebUI, an advanced web application that extends the capabilities of the DEVS-BLOOM framework, providing a user-friendly interface that supports different user roles. The application includes an interface to manage users and permissions, dashboards to inspect data (from sensors, Unmanned Surface Vehicles (USVs), weather stations, etc.), train AI models, explore their predictions, and facilitate decision-making through notification of early warnings. A key feature of DEVS-BLOOM-WebUI is the Scenario Configuration Editor (SCE). This interactive tool allows for users to design and configure the deployment of monitoring infrastructure within a water body, enhancing the system’s adaptability to user-defined simulation scenarios. This paper also investigates the practical implementation of an IoT-based EWS, discussing design considerations, sensor technologies, and communication protocols essential for seamless data integration and effective operation of the EWS.

Warming surface and Lake heatwaves as key drivers to harmful algal Blooms: A case study of Lake Dianchi, China

Extreme heat events such as marine and atmospheric heatwaves, have catastrophic impacts on marine and terrestrial ecosystems. Recent studies have shown that the continuous warming of lake surfaces and enhancement of lake heatwaves (LHWs) have generated great research interest globally. However, the effects of LHWs on lake ecosystems, especially cyanobacterial harmful blooms (cHABs), remain unclear. This study reconstructed the daily Lake surface water temperature (LSWT) time series in Lake Dianchi from 1951 to 2020 using random forest. Trends and characteristics of LSWT and LHWs were identified, and their critical driving effects on cHABs were analyzed. Results showed that the LSWT in Lake Dianchi significantly increased with a rate of 0.26 °C/10a and with an abrupt changing point in 1992, coinciding with the first report of severe cHABs in Lake Dianchi. From 1951 to 2020, 141 LHWs were detected, of which 94.3 % occurred after 1992. Among all the potential climatic and water quality driving factors on Chlorophyll-a (Chla) variation, LSWT is the most important. Chla concentration and the intensity of LHWs were significantly correlated. The complex and strong interactions between LHWs and cHABs enable the suppression of cHABs only when both remain low. A case study in Lake Dianchi highlighted the critical driving effects of LSWT and LHWs on cHABs, suggesting that continuously warming surface and enhancing LHWs in the future will further challenge more specific and effective lake management strategies to suppress cHABs.

Employing hybrid deep learning for near-real-time forecasts of sensor-based algal parameters in a Microcystis bloom-dominated lake

Harmful cyanobacterial blooms (CyanoHABs) are increasingly impacting the ecosystem of lakes, reservoirs and estuaries globally. The integration of real-time monitoring and deep learning technology has opened up new horizons for early warnings of CyanoHABs. However, unlike traditional methods such as pigment quantification or microscopy counting, the high-frequency data from in-situ fluorometric sensors display unpredictable fluctuations and variability, posing a challenge for predictive models to discern underlying trends within the time-series sequence. This study introduces a hybrid framework for near-real-time CyanoHABs predictions in a cyanobacterium Microcystis-dominated lake - Lake Dianchi, China. The proposed model was validated using hourly Chlorophyll-a (Chl a) concentrations and algal cell densities. Our results demonstrate that applying decomposition-based singular spectrum analysis (SSA) significantly enhances the prediction accuracy of subsequent CyanoHABs models, particularly in the case of temporal convolutional network (TCN). Comparative experiments revealed that the SSA-TCN model outperforms other SSA-based deep learning models for predicting Chl a (R2 = 0.45–0.93, RMSE = 2.29–5.89 μg/L) and algal cell density (R2 = 0.63–0.89, RMSE = 9489.39–16,015.37 cells/mL) at one to four steps ahead predictions. The forecast of bloom intensities achieved a remarkable accuracy of 98.56 % and an average precision rate of 94.04 % ± 0.05 %. In addition, scenarios involving various input combinations of environmental factors demonstrated that water temperature emerged as the most effective driver for CyanoHABs predictions, with a mean RMSE of 2.94 ± 0.12 μg/L, MAE of 1.55 ± 0.09 μg/L, and R2 of 0.83 ± 0.01. Overall, the newly developed approach underscores the potential of a well-designed hybrid deep-learning framework for accurately predicting sensor-based algal parameters. It offers novel perspectives for managing CyanoHABs through online monitoring and artificial intelligence in aquatic ecosystems.

Field and laboratory studies of fluorescence-based technologies for real-time tracking of cyanobacterial cell lysis and potential microcystins release

Elevated levels of dissolved microcystins (MCs) in source water due to rapid cell lysis of harmful cyanobacterial blooms may pose serious challenges for drinking water treatment. Catastrophic cell lysis can result from outbreaks of naturally-occurring cyanophages – as documented in Lake Erie during the Toledo water crisis of 2014 and in 2019, or through the application of algaecides or water treatment chemicals. Real-time detection of cyanobacterial cell lysis in source water would provide a valuable tool for drinking water plant and reservoir managers. In this study we explored two real-time fluorescence-based devices, PhycoSens and PhycoLA, that can detect unbound phycocyanin (uPC) as a potential indication of cell lysis and MCs release. The PhycoSens was deployed at the Low Service pump station of the City of Toledo Lake Erie drinking water treatment plant from July 15 to October 19, 2022 during the annual cyanobacteria bloom season. It measured major algal groups and uPC in incoming lake water at 15-min intervals during cyanobacteria dominant and senescence periods. Intermittent uPC detections from the PhycoSens over a three-month period coincided with periods of increasing proportions of extracellular MCs relative to total (intracellular and extracellular) MCs, indicating potential for uPC use as an indicator of cyanobacterial cell integrity. Following exposures of laboratory-cultured MCs-producing Microcystis aeruginosa NIES-298 (120 μg chlorophyll/L) to cyanophage Ma-LMM01, copper sulfate (0.5 and 1 mg Cu/L), sodium carbonate peroxyhydrate (PAK® 27, 6.7 and 10 mg H2O2/L), and potassium permanganate (2.5 and 4 mg/L), appearance of uPC coincided with elevated fractions of extracellular MCs. The PhycoLA was used to monitor batch samples collected daily from Lake Erie water exposed to algaecides in the laboratory. Concurrence of uPC signal and surge of dissolved MCs was observed following 24-h exposures to copper sulfate and PAK 27. Overall results indicate the appearance of uPC is a useful indicator of the onset of cyanobacterial cell lysis and the release of MCs when MCs are present.

Feasibility and mechanism of removing Microcystis aeruginosa and degrading microcystin-LR by dielectric barrier discharge plasma

Harmful cyanobacterial bloom is one of the serious environmental problems worldwide. Microcystis aeruginosa is a representative harmful alga in cyanobacteria bloom. It is of great significance to develop new technologies for the removal of Microcystis aeruginosa and microcystins. The feasibility and mechanism of removing microcystis aeruginosa and degrading microcystins by dielectric barrier discharge (DBD) plasma were studied. The suitable DBD parameters obtained in this study are DBD (41.5 W, 40 min) and DBD (41.5 W, 50 min), resulting in algae removal efficiency of 77.4% and 80.4%, respectively; scanning electron microscope and LIVE-DEATH analysis demonstrate that DBD treatment can disrupt cell structure and lead to cell death; analysis of elemental composition and chemical state indicated that there are traces of oxidation of organic nitrogen and organic carbon in microcystis aeruginosa; further intracellular ROS concentration and antioxidant enzyme activity analysis confirm that DBD damage microcystis aeruginosa through oxidation. Meanwhile, DBD can effectively degrade the microcystin-LR released after cell lysis, the extracellular microcystin-LR concentration in the DBD (41.5 W) group decreased by 88.7% at 60 min compared to the highest concentration at 20 min; further toxicity analysis of degradation intermediates indicated that DBD can reduce the toxicity of microcystin-LR. The contribution of active substances to the inactivation of microcystis aeruginosa is eaq− >•OH > H2O2 > O3 > 1O2 >•O2− >ONOO−, while on the degradation of microcystin-LR is eaq− >•OH > H2O2 > O3 >•O2− >1O2 > ONOO−. The application of DBD plasma technology in microcystis aeruginosa algae removal and detoxification has certain prospects for promotion and application.

Spatial and Temporal Variability of Saxitoxin-Producing Cyanobacteria in U.S. Urban Lakes.

Harmful cyanobacterial blooms (HCBs) are of growing global concern due to their production of toxic compounds, which threaten ecosystems and human health. Saxitoxins (STXs), commonly known as paralytic shellfish poison, are a neurotoxic alkaloid produced by some cyanobacteria. Although many field studies indicate a widespread distribution of STX, it is understudied relative to other cyanotoxins such as microcystins (MCs). In this study, we assessed eleven U.S. urban lakes using qPCR, sxtA gene-targeting sequencing, and 16S rRNA gene sequencing to understand the spatio-temporal variations in cyanobacteria and their potential role in STX production. During the blooms, qPCR analysis confirmed the presence of the STX-encoding gene sxtA at all lakes. In particular, the abundance of the sxtA gene had a strong positive correlation with STX concentrations in Big 11 Lake in Kansas City, which was also the site with the highest quantified STX concentration. Sequencing analysis revealed that potential STX producers, such as Aphanizomenon, Dolichospermum, and Raphidiopsis, were present. Further analysis targeting amplicons of the sxtA gene identified that Aphanizomenon and/or Dolichospermum are the primary STX producer, showing a significant correlation with sxtA gene abundances and STX concentrations. In addition, Aphanizomenon was associated with environmental factors, such as conductivity, sulfate, and orthophosphate, whereas Dolichospermum was correlated with temperature and pH. Overall, the results herein enhance our understanding of the STX-producing cyanobacteria and aid in developing strategies to control HCBs.

Shifts in phytoplankton and zooplankton communities in three cyanobacteria-dominated lakes after treatment with hydrogen peroxide

Cyanobacteria can reach high densities in eutrophic lakes, which may cause problems due to their potential toxin production. Several methods are in use to prevent, control or mitigate harmful cyanobacterial blooms. Treatment of blooms with low concentrations of hydrogen peroxide (H2O2) is a promising emergency method. However, effects of H2O2 on cyanobacteria, eukaryotic phytoplankton and zooplankton have mainly been studied in controlled cultures and mesocosm experiments, while much less is known about the effectiveness and potential side effects of H2O2 treatments on entire lake ecosystems. In this study, we report on three different lakes in the Netherlands that were treated with average H2O2 concentrations ranging from 2 to 5 mg L−1 to suppress cyanobacterial blooms. Effects on phytoplankton and zooplankton communities, on cyanotoxin concentrations, and on nutrient availability in the lakes were assessed. After every H2O2 treatment, cyanobacteria drastically declined, sometimes by more than 99%, although blooms of Dolichospermum sp., Aphanizomenon sp., and Planktothrix rubescens were more strongly suppressed than a Planktothrix agardhii bloom. Eukaryotic phytoplankton were not significantly affected by the H2O2 additions and had an initial advantage over cyanobacteria after the treatment, when ample nutrients and light were available. In all three lakes, a new cyanobacterial bloom developed within several weeks after the first H2O2 treatment, and in two lakes a second H2O2 treatment was therefore applied to again suppress the cyanobacterial population. Rotifers strongly declined after most H2O2 treatments except when the H2O2 concentration was ≤ 2 mg L−1, whereas cladocerans were only mildly affected and copepods were least impacted by the added H2O2. In response to the treatments, the cyanotoxins microcystins and anabaenopeptins were released from the cells into the water column, but disappeared after a few days. We conclude that lake treatments with low concentrations of H2O2 can be a successful tool to suppress harmful cyanobacterial blooms, but may negatively affect some of the zooplankton taxa in lakes. We advise pre-tests prior to the treatment of lakes to define optimal treatment concentrations that kill the majority of the cyanobacteria and to minimize potential side effects on non-target organisms. In some cases, the pre-tests may discourage treatment of the lake.
